Is glutathione a peptide?

The short answer

Yes. Glutathione is a tripeptide, a molecule made from three amino-acid units: glutamate, cysteine, and glycine. Its bond between glutamate and cysteine uses glutamate's side chain. That unusual bond still fits the chemical definition of a peptide.

Why glutathione is a peptide

IUPAC defines peptides as compounds made when amino-carboxylic-acid molecules join through a bond between a carbonyl carbon and a nitrogen atom. The definition covers more than one type of amino acid arrangement.

PubChem identifies glutathione as a tripeptide. Its three amino-acid units are glutamate, cysteine, and glycine. The NCBI Bookshelf also names it gamma-glutamyl-cysteinyl-glycine.

Tripeptide means three amino-acid units. A short peptide can still have important roles in cells. Chain length alone does not show what a finished product does in a person.

What is unusual about its structure?

Many peptides use a bond from an amino acid's alpha carboxyl group. Glutathione uses glutamate's gamma carboxyl group to join glutamate to cysteine. The final unit, glycine, joins cysteine through a peptide bond.

That gamma bond makes glutathione structurally unusual. It does not move glutathione into a different chemical class. The IUPAC definition includes compounds derived from amino-carboxylic acids more broadly.

Chemical names can make this look harder than it is. The sequence gamma-glutamyl-cysteinyl-glycine names the three connected units and the unusual first connection.

How do GSH and GSSG relate?

GSH is the reduced form of glutathione. GSSG is glutathione disulfide, formed when two glutathione molecules connect through a disulfide bond. These names describe a chemical state and do not replace the peptide classification.

The reduced and oxidized forms take part in cellular redox reactions. The NCBI Bookshelf describes glutathione as a coreactant in glutathione peroxidase and glutathione transferase reactions.

A cellular role does not establish a health outcome for a product. Evidence about a product needs the exact formulation, route, population, outcome, and study period.
